Job: Phenotypic Drug Discovery with High-Content Imaging

  • PDD with image-based high-content screening/analysis (HCS/A) is a great way of identifying new drugs based on knowledge of distinct phenotypes.
  • Cells and tissues are transparent: staining or fluorescent labeling increase cost/effort/time and may interfere with cellular function, skew the assays.
  • Fluorescence imaging (preferred) is slower and more expensive than brightfield, especially at the high magnifications required for cellular analysis.
  • Assays are much thicker than the depth of field: image multiple Z planes

  • High-resolution label-free microscopy of cells, closest to their natural state.
  • Conventional DIC and phase-contrast are not quantitative and current quantitative phase microscopy (QPM) is limited in resolution and sensitivity (animal cells/tissues have very low index contrast features, <0.1).
  • High-throughput high-resolution imaging of thick samples (cellular assays): current techniques are too slow or unreliable.

The quantitative phase, amplitude and polarization data enables reliable automated classification of cellular phenotypes without interference from stains or fluorescent labels.  Through quantitative normalization, data from different instruments/sites can be quantitatively corroborated.
